It is time to update my Computer monitor. I am looking at this one. Samsung CF398 Series Curved 27-Inch FHD Monitor. I have one question is there any functional difference between a Display Port, and a HDMI input. My Radeon 380 R9 graphics card has both.

My desktop has a 27" monitor driven by an Nvidea GTX-680. It has all three port types and so does my QNix 27" monitor. I run the RGB out to my monitor (running at 2560 x 1440 resolution). I've tried the display port and the HDMI connections. They didn't (IMO) look any better than the RGB connection. My 50" TV is directly connected to the same card via an HDMI connection and it also looks very good.

Since you have the option of using several inputs I suggest trying them all and decide for yourself.

also check your graphics card output, make sure it can send out the resolution of your new monitor.
mine has all three ports but max resolution on all three ports is 1920 x 1080 so the newer
monitors forces a graphics card upgrade.

The Way I have the current setup is the HDMI port goes to my Onkyo AVR for the excellent AMD True Audio, and the LG HDTV becomes a big screen monitor. The dedicated monitor is connected with the DVI cable.

To answer your question, no, there is no functional difference between HDMI and Display Port. They are essentially the same just different connectors. DVI is actually the same as well except it doesn't usually carry audio. RGB (or VGA in the PC world) as mentioned above is analog where as the others are digital so whether RGB or Digital looks better is hard to say and largely dependent on the specifics of the devices in question. All of those formats have enough capability to handle anything you throw at it and look just as good as any other.
